byk gak orang tanye, “ko buat honours ek chad?? ape ko buat??”.. adeh2.. meh sini aku nk habaq mai.. project hons aku bertajuk “Species and strain specific reverse transcription polymerase chain reaction (RT-PCR) detection of Wheat streak mosaic virus (WSMV) and its variants”.. basically, aku tgh develop method utk detect WSMV infection dlm host plants.. ye la, tak sumer infected plants tunjukkan symptoms, tapi it doesn’t mean die tak infected kan.. so kalo kite tak tau virus tuh ader dlm plant tuh, nnt kalo kite biar je, nnt infection tu akan spread la.. pasal tu la aku kne develop detection method ni, specific utk detect WSMV, as a first step to contain the infection..

detection method yang aku develop tu senang je.. kepada sesiapa yang tak tau.. RT-PCR method ni dh mmg ader dah.. mamat yang develop PCR ni pon dah dpt Nobel Prize dh pon.. PCR ni basically kite gune primer or oligonucteotide (short sequences of DNA) utk amplify specific target region.. well, setiap hidupan kt muke bumi ni ade DNA or RNA sendiri yang sgt unique.. so what i have to do is to design primers that are specific to WSMV sahaje.. tapi PCR ni leh wat kt DNA je and since WSMV is a RNA virus, so firstly aku kene la wat RT so that leh dpt cDNA (complementary DNA; single strand DNA yang diprepare dari RT of RNA)..

okeh, tgk title project aku, ade 2 parts kan?? satu species specific, and another one is strain specific.. WSMV tu name species virus yang aku nk detect.. but still, dlm setiap species, ade variants (sub-species).. variants ni wujud due to mutations.. survival of the fittest la.. ade yang mutate camni, ade yang cam gtu.. mane yang survive mutation die, die akan replicate, creating a new sub-species.. so, over time, ade gak arr beberapa sub-species yang survive.. every sub-species ni ade gak differences dlm DNA sequences diorang.. so, aku nk develop a method nk differentiate polymorphisms dlm variants ni gak arr.. nk pakai PCR boley, tapi tak practical sgt.. coz tak byk pon sgt differences diorg.. so aku kene gune method yang leh differentiate even 1 point mutation.. i’m thinking of using restriction fragment length polymorphisms (RFLP).. utk RFLP method ni kite gune restriction enzyme untuk cut at specific region dlm DNA sequence.. so, for example kite ade 2 variants, salah satu variant tu ade mutation kt restriction site tu, maka restriction enzyme tu xleh la cut variant tu.. so nnt kite leh arr tgk restriction pattern yang dihasilkan and tau, ok this 2 samples are 2 different variants.. nice eh??
